EFFECT OF BORATE ON A PROTEIN-POLYSACCHARIDE COMPLEX, THE PHOSPHOESTERASE FROM CALF INTESTINAL MUCOSA sirs: An enzyme preparation from calf intestinal mucosa

چکیده

An enzyme preparation from calf intestinal mucosa was studied for phosphodiesterase activity’ but was found to contain also phosphomonoesterase’ and adenosine deaminase.2 Analysis confirmed the presence of polysaccharide, suspected because of solubility in high concentrations of organic solvents and precipitation from aqueous solutions only when fully saturated with ammonium sulfate. Attempts were made to obtain a more active phosphodiesterase fraction with little success. It was concluded that enzymes and inert proteins had closely similar properties, perhaps all being protein-polysaccharide complexes.

Adenosine deaminase from calf intestinal mucosa.

In the course of studying the phosphoric acid esterases from calf intestinal mucosa (1, 2) the preparations were found to contain a potent deaminase for adenosine. The method of preparation is simple and the procedure indicates that this enzyme is quite stable. This deaminase is probably identical with that recently prepared by Brady (3) from the same material by a somewhat different procedure....

Affinity-chromatography purification of alkaline phosphatase from calf intestine.

A crude preparation of alkaline phosphatase (EC 3.1.3.1) from calf intestinal mucosa was purified by affinity chromatography on Sepharose-bound derivatives of arsanilic acid, which was found to be a competitive inhibitor of the enzyme. Three biospecific adsorbents were prepared for the chromatography, and the best results were obtained with a tyraminyl-Sepharose derivative coupled with the diaz...

Identification of diagnostic biomarkers by bioinformatics analysis in the inflamed and non-inflamed intestinal mucosa in Crohn\'s disease patients

Background: Crohn's disease (CD) is a type of inflammatory bowel disease (IBD) which despite the unknown details is generally related to genetic, immune system, and environmental factors. In this study, we identify transcriptional signatures in patients with CD and then explain the potential molecular mechanisms in inflamed and non-inflamed intestinal mucosa in these patients. Materials and Me...

An Alkaline Phosphatase Lacking Wheat Germ Agglutinin Binding Sites Useful Enzyme for Lectin Assays with Comparable Activity to the Calf Enzyme

Despite the availability of various alkaline phosphatase (ALP) isoenzymes, the calf enzyme is being used in current enzyme assays as the detector enzyme. The glycosylation pattern of this enzyme makes it a suitable ligand for binding to wheat germ agglutinin lectin (WGA). As a result of this property, the enzyme can not be used as a conjugate with this lectin, and the calf enzyme conjugates can...

Bacterial Expression and Functional Characterization of A Naturally Occurring Exon6-less Preprochymosin cDNA

Chymosin (Rennin EC 3.4.23.4), an aspartyl proteinase, is the major proteolytic enzyme in the fourthstomach of the unweaned calf, and it is formed by proteolytic activation of its zymogene, prochymosin.Following the cloning of synthesized cDNAs on mRNA pools extracted from the mucosa of the calf fourthstomach, we have identified an alternatively spliced form of preprochymosin ...
